Review finds no causal link between prenatal acetaminophen and neurodevelopmental disorders

A comprehensive “umbrella” review published in Developmental Medicine & Child Neurology has concluded that existing scientific evidence does not substantiate a causal link between prenatal exposure to the pain reliever paracetamol (acetaminophen) and neurodevelopmental disorders such as autism spectrum disorder and attention‑deficit/hyperactivity disorder (ADHD). The analysis pooled data from multiple prior investigations, many of which were limited by small sample sizes, methodological shortcomings, or reported only modest associations between maternal paracetamol use and later child outcomes.

The review highlighted that while earlier studies have suggested possible connections, the overall body of research lacks the robustness required to infer causality. Researchers emphasized that the current evidence base is insufficient to support definitive claims that prenatal paracetamol exposure directly contributes to the development of autism or ADHD. Consequently, clinicians and pregnant individuals are advised to consider the broader context of existing data when evaluating the safety of paracetamol use during pregnancy.
